Research Progress on the Combination of ADC Drugs and Immunotherapy in Non-Small Cell Lung Cancer

Zhuocheng Peng

Abstract

Globally, lung cancer is the most common cause of cancer mortality and most patients have non-small cell lung cancer (NSCLC),
which has a poor prognosis. Molecularly targeted drugs and immune checkpoint inhibitors (ICIs) have become the mainstay of first-line treatment, but drug resistance significantly affects the durability of clinical responses. However, the combination of ADCs with ICIs may have
synergistic potential as ADCs not only mediate targeted tumor killing but also have immunomodulatory properties such as immunogenic cell
death and activation of dendritic cells. However, achieving durable remission, even after the current progress with targeted therapy, anti-angiogenic agents, ICIs and ADCs, is a huge challenge. The goal of this review is to provide an overview on the synergistic mechanisms acting in
ADC–immunotherapy combination approaches, recent clinical advances in first-line treatment, and safety management considerations to help
guide clinical practice and future investigations.
