A Mechanistic Evaluation of the Effects of Astragaloside IV on Neurite Outgrowth Under Oxygen-Glucose Deprivation Conditions

Abstract


This study examined the effects of Astragaloside IV (AS-IV) on neurite outgrowth in PC12 cells subjected to oxygen-glucose deprivation (OGD). Cell viability assays demonstrated that increased OGD duration led to reduced cell viability, neurite length, and branching. ASIV treatment (20, 40, 80, and 160 g/ml) reversed these effects, enhancing cell activity and promoting longer neurites. Immunohistochemistry
showed elevated RhoA expression and decreased levels of growth-promoting proteins Rac1 and Cdc42 following OGD, but AS-IV treatment
lowered RhoA and increased Rac1 and Cdc42 levels. Additionally, pull-down assays indicated that AS-IV mitigated the activation of RhoA
while enhancing Rac1 and Cdc42, thus supporting neurite outgrowth by preventing collapse and promoting growth cone expansion.
