Bidirectional Causal Association Between Chronic Obstructive Pulmonary Disease and Chronic Periodontitis: A Two-sample Mendelian Randomization Study

Yanchen Yang, Zehui Li*

Abstract

Objective: The purpose of this study was to investigate whether there is a causal relationship between Periodontitis and Chronic
Obstructive Pulmonary Disease by Mendelian randomization analysis. Method: We performed a two-sample Mendelian randomization
analysis using publicly released genome-wide association studies (GWAS) statistics. The inverse-variance weighted method was used as
the primary analysis. Result: IVW analysis showed no effect of Periodontitis on COPD. Similarly, no causal relationship between COPD
and Periodontitis was found in reverse analysis. The results of MR-Egger regression, weighted median, and weighted mode methods
were consistent with those of the IVW method. Based on sensitivity analyses, horizontal pleiotropy is unlikely to distort causal estimates.
Conclusion: Although observational studies have reported an association between Periodontitis and COPD, the results of our analysis do
not support a causal relationship between them. Clinical relevance MR study can analyze the causal relationship between Periodontitis and
COPD, to provide a certain reference for clinicians and deepen the understanding of the relationship between Periodontitis and COPD.
