The Effect of Free Bone Flap Suspension on the Expression of IL-10 in Rabbits
Abstract
IL-10 in rabbit brain tissue and to provide a theoretical basis for postoperative inflammation control and tissue repair. Methods: Thirty New
Zealand white rabbits were randomly divided into suspension group, non-suspension group and control group to establish craniocerebral injury models or craniotomy alone. ELISA, immunohistochemistry and qPCR were used to detect serum IL-10 concentration and IL-10 protein
and mRNA expression levels in brain tissue on the 1st, 3rd and 7th days after surgery, and general physiological state and survival rate were
observed. Results: On the third day after surgery, the serum IL-10 concentration in the suspension group rose to (65.7 8.9)pg/mL, significantly higher than that in the non-suspension group (42.3 6.5)pg/mL and the control group (29.4 5.0)pg/mL (P<0.01). The immunohistochemical score of the suspension group was 5.6 0.8, higher than that of the non-suspension group (3.2 0.9), and IL-10 mRNA expression
was also significantly upregulated (1.82 0.37 vs 1.15 0.28). Correlation analysis showed that IL-10 mRNA was positively correlated with
protein expression (r = 0.85) and negatively correlated with the degree of inflammatory cell infiltration (r = -0.79). Conclusion: Free bone flap
suspension can effectively increase IL-10 expression, reduce local inflammatory response after surgery, and has certain neuroprotective and
immunomodulatory effects.
