The Analysis of Effect of Bushen Huayu Formula on the NLRP3/ASC/Caspase-1 Signaling Pathway in Ovarian Tissue of Infertile Rats with Kidney Deficiency and Blood Stasis

Abstract


This study investigates the therapeutic effects of Bushen Huayu Formula (BHF) on ovarian tissue in infertile rats with kidney deficiency and blood stasis syndrome, focusing on the NLRP3/ASC/Caspase-1 signaling pathway. Forty-eight female Sprague-Dawley rats were
randomly divided into four groups: control, model, BHF-treated, and positive control. The kidney deficiency and blood stasis model was
established using hydroxyurea and adrenaline injections combined with cold stress. BHF administration significantly improved ovarian morphology, reduced inflammatory factors, and downregulated the expression of NLRP3, ASC, and Caspase-1 proteins compared to the model
group. Quantitative analysis revealed that BHF treatment decreased NLRP3 expression by 42.7% (p 0.01) and Caspase-1 activity by 38.2% (p
0.05). These findings suggest that BHF ameliorates ovarian dysfunction by modulating the NLRP3 inflammasome pathway, providing potential therapeutic targets for female infertility associated with kidney deficiency and blood stasis.
