Establishment and Validation of Multiplex PCR Assay for Simultaneous Identification of Four Common Foodborne Pathoge

Abstract


This study established and optimized a quadruple multiplex PCR assay to simultaneously detect Salmonella, Staphylococcus aureus,
Listeria monocytogenes and Escherichia coli O157: H7. It had good specificity with no cross-reaction to non-target bacteria. The detection
limits were very low for pure cultures and also low for artificially contaminated food after pre-enrichment. The assay showed excellent repeatability. For a large number of commercial food samples, its consistency with the national standard method was very high. The detection time
was greatly shortened to a short period, with high efficiency, low cost and strong anti-interference.
